Dihydroethidium (DHE) is a peroxide indicator that penetrates cell membranes to form a fluorescent protein complex emitting blue fluorescence. Inside cells, Dihydroethidium mainly localizes in the cell membrane, cytoplasm, and nucleus, with the strongest staining effect in the nucleus. It exhibits inherent blue fluorescence with a maximum excitation wavelength of 370 nm and a maximum emission wavelength of 420 nm. After dehydrogenation, it combines with RNA or DNA to produce red fluorescence with a maximum excitation wavelength of 300 nm and a maximum emission wavelength of 610 nm, with 535 nm also usable for excitation during observation.

Description
Dihydroethidium (DHE) is a peroxide indicator that readily penetrates cell membranes and forms a fluorescent protein complex. It predominantly accumulates in the cell membrane, cytoplasm, and nucleus, with the most intense staining observed in the nucleus. Dihydroethidium naturally emits blue fluorescence, with a peak excitation wavelength at 370 nm and emission at 420 nm. Upon dehydrogenation, it binds to RNA or DNA to produce red fluorescence, peaking at 300 nm for excitation and 610 nm for emission. An excitation wavelength of 535 nm is also applicable for practical observation[1].
