With dicyclohexyl-carbodiimide; In dichloromethane; at 20℃; To a stirred solution of 2,8-dimethyl-l-thia-3,8-diaza-spiro[4.5]decane (Example 1) (0.83 g, 44 mmol) in dichloromethane (85 ml) at room temperature was added dicyclohexylcarbodiimide (DCC) (1.23 g, 5.9 mmol) followed by addition of trans-3-indoleaceticrylic acid (1.0 g, 5.3 mmol). The resulting solution was stirred at room temperature overnight. During the reaction a white solid has precipitated. After filtration the solvent was evaporated and the crude product was purified by flash chromatography (silica, CH2Cl2/ MeOH /NH4OH 90/10/1) to give the title compound (600 mg) as a solid. 1HNMR (l,l,2,2-Dichloroethane-d2, 300 MHz, 1000C) delta 8.56 (br s, IH, NH-indole), 7.94 (d, J= 15.3 Hz, IH, HC=C), 7.83 (m, IH, CHC arom), 7.46 (br s, IH, CHC arom), 7.42 (m, IH, CH), 7.26 (m, 2H, 2CH arom), 6.71 (d, J= 15.3 Hz, IH, C=CH), 5.65 (q, J= 6.12 Hz, IH, CHCH3), 4.38 (d, J= 11.75 Hz, IH, CHHNCO), 3.50 (d, J= 11.84 Hz, IH, CHHNCO)5 3.1-2.8 (m, 2Eta), 2.8-2.6 (m, IH), 2.6-2.3 (m, 2H), 2.46 (s, 3H, NCH3), 2.2-1.9 (m, 2H), 1.9-1.8 (m, IH), 1.64 (d, J= 6.21 Hz, 3H, CH3-CH) ppm; MS (EI+) m/z 355 (M+), 322, 185, 170.
