With triethylamine; In dichloromethane; ethyl acetate; Preparation 45 N-[(1,4-Dichloro-7-isoquinolinyl)sulphonyl]-L-proline t-butyl ester STR148 A mixture of L-proline t-butyl ester hydrochloride (335 mg, 1.61 mmol), NEt3 (0.53 mL, 3.78 mmol) and 1,4-dichloro-7-isoquinolinesulphonyl chloride (449 mg, 1.51 mmol) in CH2 Cl2 (10 mL) was stirred at 23 C. for 20 h. The solvents were evaporated in vacuo and the residue suspended in EtOAc. This solution was washed with water, brine, dried (MgSO4) and evaporated in vacuo. The residue was purified by column chromatography upon silica gel using pentane-EtOAc (90:10 to 70:30) as eluant to give N-[(1,4-dichloro-7-isoquinolinyl)sulphonyl]-L-proline t-butyl ester (543 mg, 1.26 mmol) as a white solid. 1 H (CDCl3, 300 MHz) delta 1.45 (9H, s), 1.8-2.1 (3H, m), 2.1-2.3 (1H, m), 3.4-3.6 (2H, m), 4.4 (1H, dd), 8.3 (1H, d), 8.4 (1H, d), 8.5 (1H, s), 8.9 (1H, d) ppm. LRMS 431 (MH+), 448, 450 (MNH4+). Anal. Found: C, 50.09; H, 4.62; N, 6.37. Calc for C18 H20 Cl2 N2 O4 S: C, 50.12; H, 4.67; N, 6.49.
