YieldReaction ConditionsOperation in experiment
84% With N-ethyl-N,N-diisopropylamine; In isopropyl alcohol; at 90℃; for 5h; A solution of [349-02-0]4-fluoro-3-nitrobenzamide (50 mg, 1.3 mmol) ini-PrOH (5 ml) was treated with DIPEA (0.14 ml, 3.8 mmol) and (R)-4-aminopropan-2-ol (0.06 ml, 3.8 mmol). After stirring at 90oC for 5 h, solvent was removed under reduced pressure to give a crude product which was purified by column chromatography (SiO2, CH2Cl2:MeOH:NH4OH:H2O = 80:20:1:1) to give5cas a yellow powder (52 mg, 0.22 mmol, 80% yield);1H NMR (400 MHz, (CD3)2SO) delta (ppm) 8.66 (s, 1H), 8.49 (t,J= 4.7 Hz, 1H), 8.00 (d,J= 8.8 Hz, 1H), 7.99 (s, 1H), 7.28 (br s, 1H), 7.12 (d,J= 9.1 Hz, 1H), 5.06 (d,J= 4.9 Hz, 1H), 3.64 (br s, 1H), 3.45 - 3.40 (m, 1H), 3.21 - 3.17 (m, 1H), 1.16 (d,J= 6.2 Hz, 3H).
