The reaction was filtered and the filtrate was concentrated to yield crude 1-iodo-2-(2-iodoethoxy)ethane.To a mixture of 1-iodo-2-(2-iodoethoxy)ethane (848 mg, 2.6 mmol), LHMDS (5 mL, 1.0 M), and THF (5 mL) was added a solution of methyl 1-benzyl-4-(cyanomethyl)-1H-pyrrole-2-carboxylate (500 mg, 2.0 mmol) in THF (2.5 mL). The reaction was complete after stirring at room temperature for 15 min. An aqueous solution of saturated ammonium chloride was added followed by ethyl acetate and water. The mixture was extracted with ethyl acetate (1×) and the combined organic extracts were washed with brine (1×), dried over magnesium sulfate, and concentrated. The crude product was purified by chromatography on silica gel (0-50% ethyl acetate/hexane) to yield the pure methyl 1-benzyl-4-(4-cyanotetrahydro-2H-pyran-4-yl)-1H-pyrrole-2-carboxylate. MS: (M+H)+=324.8. |
