66% With dmap; 1-ethyl-(3-(3-dimethylamino)propyl)-carbodiimide hydrochloride; In N,N-dimethyl-formamide; at 45℃; for 24h; General procedure: A solution of benzyl aminobenzoate 9a (1.20 g, 5.28 mmol) in dry DMF (7 mL) was treated with 4-(dimethylamino)butanoic acid hydrochloride 18a13 (1.77 g, 10.6 mmol), EDC (1.64 g, 10.6 mmol) and DMAP (1.29 g, 10.6 mmol) and the mixture was stirred at 45 C for 24 h. The reaction mixture was then diluted with 1:1 DCM/isopropanol (60 mL), washed with saturated aqueous sodium bicarbonate (50 mL), dried (MgSO4), filtered and the solvent was removed in vacuo. The crude product was dissolved in methanol (4 mL), silica gel was added until a consistent slurry was formed. The solvent was removed to give the crude product adhered to silica. This silica gel was then loaded and purified by flash chromatography (CH2Cl2/MeOH 19:1) to give the title compound 19a (1.17 g, 65%) as a clear oil.
