66.5 grams (93%) With pyridine; In water; acetone; EXAMPLE A Preparation of Bis(o-carboxyphenyl)succinate 0.4 moles salicylic acid (55.2 grams) 0.4 moles pyridine (31.2 ml.) 0.2 moles succinyl chloride (31.0 grams) The salicylic acid and pyridine are dissolved in 90 ml. of acetone. To the resulting clear solution, the succinyl chloride in 90 ml. of acetone is added, with stirring, at a rate to keep acetone refluxing for half an hour. During this addition, the desired BOCS product begins to separate and the mixture turns dark purple. The reaction slurry is stirred for half an hour after all the succinyl chloride has been added. Then 200 ml. of water are added, the acetone evaporated from the slurry in a rotary evaporator at 35 C. and the desired BOCS product collected on a suction filter, washed with water and dried in vacuum at 60 C. Yield--66.5 grams (93%). M.P. 177-177.5 C.
With pyridine; In tetrahydrofuran; 1. The compound of Formula (I) wherein Y represents -C(=0)-X and X represents CH2CH2- substituted with the following structure by attachment through the carbon the -C(0)0 group of the structureand R and Rl represent -OH,[bis(2-carboxyphenyl) succinate] or BCS, example compound no. 508 in table 1 , was synthesised by the following method:Salicylic acid (27.6g; 0.2mol) was added to a 2 neck 250 cm3 Quick-fit round bottom flask. Dry THF (75 cm3 ; dried over anhydrous magnesium sulphate for 24h) was then added followed by pyridine (16.1 ml; 0.2 mol). The flask was then fitted with a dropping funnel, condenser and silica-gel guard tubes. The solution was stirred until the salicylic acid had completely dissolved. A solution of succinyl chloride ( 1 1.0 cm3 ; O. lmol) in dry THF (20 cm3) was prepared and added slowly (over 20-30 min) with vigorous stirring. Heat was evolved and typically a purple colour developed along with a copious precipitate. Stirring was continued for a further minimum of lh, although stirring up to 18h modestly increases the yield further. The resulting mixture was then poured slowly into ice cold water (300 - 400 cm3) which was rapidly stirred in a 11 beaker. Stirring was continued (up to lh) until the precipitated product was sufficiently crystalline to be filtered with a Buchner apparatus. The crude yield was typically 29.5g (82%). The product was re-crystallised from ethanol to give a white solid with a melting point of 152- 155C; the literature value is 155- 160C (Aldrich catalogue).The reaction was stirred throughout and the stirring was mechanical stirring.
