67% With acetic acid; at 110℃;Sealed tube; Step 3. 3-(4-Fluorophenyl)-2-methyl-5-(pyridin-3-yl)pyrazolo[1,5-a]pyrimidin-7(4H)-one A mixture of 4-(4-fluorophenyl)-3-methyl-1H-pyrazol-5-amine (127 mg, 0.60 mmol, 1.00 equiv, 90%) and ethyl 3-oxo-3-(pyridin-3-yl)propanoate (116 mg, 0.60 mmol, 1.00 equiv) in acetic acid (2 mL) was placed into an 8-mL sealed tube. The resulting mixture was stirred overnight at 110 C. Then the reaction was concentrated to dryness, and the residue was suspended in 5 mL of methanol. The resulting solids were collected by filtration and washed with methanol. Drying afforded 130 g (67%) of 3-(4-fluorophenyl)-2-methyl-5-(pyridin-3-yl)pyrazolo[1,5-a]pyrimidin-7(4H)-one as a yellow solid. LC-MS: (ES, m/z): 321 [M+H]+ 1H-NMR (300 MHz, DMSO, ppm): delta 12.23 (s, 1H), 8.96 (s, 1H), 8.74 (d, J=4.8 Hz, 1H), 8.19 (d, J=6.9 Hz, 1H), 7.58 (d, J=4.8 Hz, 3H), 7.31 (t, J=8.7 Hz, 1H), 6.07 (s, 1H), 2.32 (s, 3H)
