94% With potassium hydroxide; In N,N-dimethyl-formamide; at 20℃; for 2h; To a solution of <strong>[38183-03-8]7,8-dihydroxy-2-phenyl-4H-chromen-4-one</strong> (<strong>[38183-03-8]7,8-dihydroxyflavone</strong>, 1a) (254 mg, 1 mmol) in DMF (5.0 mL) was added methyl iodide (0.25 mL, 4 mmol, 4 equiv) and K2CO3 (1.11 g, 8 mmol, 8equiv). The reaction mixture was stirred at room temperature for 2 h. DMF was evaporated in vacuo and water (30 mL) was added to the crude mixture followed by extraction with ethyl acetate (3 × 30mL). The combined organic phases were washed with brine and dried over Na2SO4 and concentrated in vacuo. The title compound was obtained after recrystallization from ethyl acetate as greenish needles (264 mg, 94%). Rf=0.65 (2% MeOH/CH2Cl2).
With potassium carbonate; In acetone; at 60℃; General procedure: Methyl iodide (25 mmol, 5 eq) was added to a solution of potassium carbonate (30 mmol, 6 eq) and flavones (5mmol, 1.0 equal) or isoflavones (5 mmol, 1.0 equal) in acetone (60 mL). The reaction mixture was stirred overnight at 60C, and the solvent was then evaporated in vacuum. The residue was purified by silica gel column chromatography, eluting with a solution of 15% MeOH in dichloromethane (Scheme S2), to yield yellow solid 15-19
