90% at 85℃; for 2 h; To a solution of Scheme 57 compound 4 (1.5 g, 6.84 mmol) in dry EtOH (15 mL) was added NaCN (362 mg, 7.53 mmol) and the reaction mixture was stirred at 85 °C for 2 h. After TLC showed the starting material was completely consumed, the reaction mixture was concentrated to give a crude compound which was diluted with ice water (30 mL) and extracted with EtOAc (2 x 50 mL). The organic layer was washed with brine (20 mL) then dried over Na2S04 and concentrated to give a residue which was purified by flash chromatography on silica gel (petroleum ether/EtOAc 100/0 gradually increasing to 70/30) to give Scheme 57 compound 5 (1 g, 90percent) as a colorless liquid. 1H NMR (400 MHz, DMSO- d6): δ 7.21 (t, J= 9.2 Hz, 1H), 7.01 (dd, J= 6.1, 3.1 Hz, 1H), 6.94 (m, 1H), 4.01 (s, 2H), 3.75 (s, 3H). MS [ESI, MH+] = 166.06.
