YieldReaction ConditionsOperation in experiment
64% Potassium f-butoxide (11.35g, 101.3mmol) was added portionwise to a solution of 3-pyrrolidin-1-ylpropan- 1-ol {Australian Journal of Chemistry, 52, 12, 1131-1138, 1999) (13.05g, 101.3mmol) in tetrahydrofuran (150ml), maintaining the reaction temperature below 3O0C. The reaction mixture was stirred at room temperature for 30 minutes. 4-(6-Chloropyridin-3-yl)tetrahydro-2H-pyran-4-carbonitrile (15g, 67.4mmol) was added portionwise and the reaction mixture stirred at room temperature for 20 hours. The mixture was diluted with diethyl ether (200ml), washed with water (100ml), brine (100ml), dried over magnesium sulphate, filtered and evaporated in vacuo. The crude material was purified by column chromatography using a Combiflash.(R). silica cartridge eluting with dichloromethane to dichloromethane: methanol: ammonia (80:20:2). The title compound was obtained as an orange oil (13.55g, 43mmol, 64percent). 1H-NMR (CDCl3, 400MHz); .delta 1.8 (m, 4H), 2.0 (m, 6H)1 2.5 (m, 4H), 2.6 (m, 2H), 3.9 (m, 2H), 4.1 (m, 2H), 4.35 (t, 2H), 6.8 (d, 1 H), 7.6 (d, 1 H), 8.25 (s, 1 H).
