43.7% | at 90℃; | Step 2: Tetrahydro-furan- 3-yl)-acetonitrile [00276] To a stirred solution of Toluene-4- sulfonic acid tetrahydro-furan-3-ylmethyl ester (17.00 g. 65.83 mmol, 1.00 eq) in dry DMF (51.45 ml, 658.33 mmol, 10.00 eq) was added Sodium Cyanide (6.47 g, 131.67 mmol, 2.00 eq). The reaction mixture was heated overnight at 90 °C, and after the completion of the reaction as confirmed by TLC, the reaction mixture was extracted with ethyl acetate (100 mL) and washed with water (3 x 25 mL). The combined organic layer was dried over anhydrous Na2S04, filtered, concentrated under vacuum. The residue was purified by column chromotography and the product was eluted with EtOAc in petroleum ether (10- 20percent) to afford (Tetrahydro-furan- 3-yl)-acetonitrile (3.20 g, 28.79 mmol, 43.7 percent) as colorless oil. lU NMR (400 MHz, DMSO-d6) δ 4.03-4.00 (m, 1H), 3.82-3.77 (m, 1H), 3.67-3.63 (m, 1H), 2.79-2.78 (m, 1H), 2.75-2.64 (m, 1H), 2.02-1.97 (m, 1H), 1.89-1.81 (m, 2H), 1.57-1.52 (m, 1H). |
